Distinct role of protein phosphatase 2A subunit Cα in the regulation of E-cadherin and β-catenin during development

چکیده
Protein phosphatase 2A (PP2A) plays central roles in development, cell growth and transformation. Inactivation of the gene encoding the PP2A catalytic subunit Ca by gene targeting generates a lethal embryonic phenotype. No mesoderm is formed in Ca embryos. Here, we found that during normal early embryonic development Ca was predominantly present at the plasma membrane whereas the highly homologous isoform Cb was localized to the cytoplasm and nuclei, suggesting the inability of Cb to compensate for vital functions of Ca in Ca embryos. In addition, PP2A was found in a complex containing the PP2A substrates E-cadherin and b -catenin. In Ca embryos, E-cadherin and b -catenin were redistributed from the plasma membrane to the cytosol. Cytosolic concentrations of b -catenin were low. Our results suggest that Ca is required for stabilization of E-cadherin/b -catenin complexes at the plasma membrane. q 2000 Elsevier Science Ireland Ltd. All rights reserved.

Beta-catenin Forms Protein Aggregation at High Concentrations in HEK293TCells
Background: The canonical Wnt signal transduction (or the Wnt/β-catenin pathway) plays a crucial role in the development of animals and in carcinogenesis. Beta-catenin is the central component of this signaling pathway. The activation of Wnt/β-catenin signaling results in the cytoplasmic and nuclear accumulation of β-catenin. In the nucleus, β-catenin interacts with the TCF/LEF transcription fa...
متن کاملInvolvement of SET in the Wnt signaling pathway and the development of human colorectal cancer
The SET oncoprotein is involved in cancer progression by modulating multiple cellular processes, including the inhibition of the tumor suppressor, protein phosphatase 2 (PP2A). Based upon these multiple activities, we hypothesized that targeted inhibition of SET is likely to have multiple discrete and measurable effects on cancer cells. In the present study, the mRNA expression levels of SET, P...
متن کاملImpaired development of the Harderian gland in mutant protein phosphatase 2A transgenic mice
Although Harderian glands are especially large in rodents, many features of this retroocular gland, including its development and function, are not well established. Protein phosphatase 2A (PP2A) is a family of heterotrimeric enzymes expressed in this gland. PP2A substrate specificity is determined by regulatory subunits with leucine 309 of the catalytic subunit playing a crucial role in the re...
متن کاملActive β-catenin is regulated by the PTEN/PI3 kinase pathway: a role for protein phosphatase PP2A
Dysregulation of Wnt/β-catenin signaling has been associated with the development and progression of many cancers. The stability and subcellular localization of β-catenin, a dual functional protein that plays a role in intracellular adhesion and in regulating gene expression, is tightly regulated. However, little is known about the transcriptionally active form of β-catenin, Active Beta Catenin...

1.Introduction Among the various tissues of the body, the heart is one of the main and most important organs of the body to which human life depends, and any tissue and physiological damage to it disrupts human health (10) . On the other hand, today, prescribing sports activities is accepted as an effective prescription in the prevention and treatment of many diseases and improving heart f...
